چکیده

I . A technique for the separation and colorimetric estimation of z,4-diaminopimelic acid (DAP) using automated ion-exchange chromatography coupled with an acid ninhydrin detection system is described. 2. Only traces of DAP were found in rumen protozoa and no DAP was detected in rumen fluid prepared by ultracentrifugation or dialysis. 3. The concentration of DAP in rumen bacteria from sheep on a constant feeding regimen, and the ratio of nitrogen to DAP for these bacteria were found to be constant over a 3-month period. 4. The method has proved suitable for the estimation of bacterial N in the duodenal digesta of ruminants. 5. The contribution of bacterial N to the total N leaving the abomasum of a lactating cow fitted with a permanent re-entrant cannula in the duodenum was found to be 5 0 % .

Determination of assay parameters for RNA analysis in bacterial and duodenal samples by spectrophotometry. Influence of sample treatment and preservation.

A simple analytical procedure derived from that described by Zinn and Owens (1980), based on the determination of nucleic purine bases (RNA eq), was carried out to measure microbial nitrogen flow in the ruminant duodenum. Several procedures for sample preservation were tested; the efficiency of each step of the analytical method was also determined using high performance liquid chromatography (...

Estimation of the proportion of bacterial nitrogen in canine feces using diaminopimelic acid as an internal bacterial marker.

A bacterial marker can be used to determine the portion of fecal N that is of bacterial origin, as well as the effect of dietary factors on the bacterial N in feces of the dog. Two experiments were conducted to determine the efficacy of diaminopimelic acid (DAPA) and purines as bacterial markers in dogs. In Exp. 1, five adult female dogs were fed the same commercial diet. In Exp. 2, 50 dogs wer...

The conversion of plant nitrogen to microbial nitrogen in the rumen of the sheep.

An estimate of the extent of conversion of plant nitrogen to microbial nitrogen in the rumen of sheep fed on wheaten hay was reported in an earlier communication (Gray & Pilgrim, 1953). The basis of separation of plant from microbial material was simply the filtration of the rumen contents through muslin with subsequent limited washing of the fibrous plant residues to free them from the remaini...

The effect of heat treatment of whole cottonseed on site and extent of protein digestion in dairy cows.

Six mature Holstein cows in late lactation and fitted with permanent ruminal, duodenal and ideal cannulae were used in a double 3 X 3 Latin-square design to study the effect of roasting or extruding of cottonseed on protein degradation in the rumen and availability of amino acids in the small intestine. Raw whole cottonseed (CS), extruded whole cottonseed (ECS), and roasted whole cottonseed (RC...
